As reported at github.com/cms-sw/cmssw/pull/49969, TClass and TStreamerInfo are unaware of (custom) alignment of classes. The consequence (seen in the report) is mis-alignment of those kind of objects when used inside an emulated classes (for example when creating the cache for the input of a read rule).
